چکیده

Messenger ribonucleoprotein (mRNP) granules are dynamic, self-assembling structures that harbor nontranslating mRNAs bound by various proteins that regulate mRNA translation, localization, and turnover. Their importance in gene expression regulation is far reaching, ranging from precise spatial-temporal control of mRNAs that drive developmental programs in oocytes and embryos, to similarly exquisite control of mRNAs in neurons that underpin synaptic plasticity, and thus, memory formation. Analysis of mRNP granules in their various contexts has revealed common themes of assembly, disassembly, and modes of mRNA regulation, yet new studies continue to reveal unexpected and important findings, such as links between aberrant mRNP granule assembly and neurodegenerative disease. Continued study of these enigmatic structures thus promises fascinating new insights into cellular function, and may also suggest novel therapeutic strategies in various disease states.

Cytoplasmic mRNP granules at a glance.

Introduction From their transcriptional birth to their degradation, cellular mRNAs are coated with proteins in messenger ribonucleoprotein (mRNP) complexes. The mRNP composition controls every aspect of the life of the mRNA, from pre-mRNA processing to mRNA localization, translation and turnover. Transitions between these events are accompanied by major mRNP remodeling and exchange of mRNP prot...

Maternal mRNAs are regulated by diverse P body–related mRNP granules during early Caenorhabditis elegans development

Processing bodies (P bodies) are conserved mRNA-protein (mRNP) granules that are thought to be cytoplasmic centers for mRNA repression and degradation. However, their specific functions in vivo remain poorly understood. We find that repressed maternal mRNAs and their regulators localize to P body-like mRNP granules in the Caenorhabditis elegans germ line. Surprisingly, several distinct types of...

P-bodies and stress granules: possible roles in the control of translation and mRNA degradation.

The control of translation and mRNA degradation is important in the regulation of eukaryotic gene expression. In general, translation and steps in the major pathway of mRNA decay are in competition with each other. mRNAs that are not engaged in translation can aggregate into cytoplasmic mRNP granules referred to as processing bodies (P-bodies) and stress granules, which are related to mRNP part...

Inhibition in cortical circuits

photobleaching reveal that the half-life of stress-granule-associated RNA-binding proteins is very brief, on the order of seconds to minutes, despite the fact that time-lapse microscopy reveals that individual stress granules persist for hours. This rapid shuttling of protein and RNA within stress granules suggests that their mRNP contents are continually sorted via fleeting associations with t...

NEDD4 controls spermatogonial stem cell homeostasis and stress response by regulating messenger ribonucleoprotein complexes

P bodies (PBs) and stress granules (SGs) are conserved cytoplasmic aggregates of cellular messenger ribonucleoprotein complexes (mRNPs) that are implicated in mRNA metabolism and play crucial roles in adult stem cell homeostasis and stress responses. However, the mechanisms underlying the dynamics of mRNP granules are poorly understood. Here, we report NEDD4, an E3 ubiquitin ligase, as a key re...
